Phosphatidylserine is a phospholipid concentrated in brain cell membranes that plays a role in cell signaling and neurotransmitter release. Several RCTs have demonstrated that PS supplementation attenuates cortisol and ACTH responses to standardized stress tests, including intense exercise and mental arithmetic challenges. The mechanism appears to involve modulation of ACTH release from the anterior pituitary, thereby reducing downstream cortisol secretion from the adrenal glands. A key study by Monteleone and colleagues showed that 800mg/day of bovine-derived PS significantly blunted the cortisol response to physical stress. However, modern supplements use soy-derived or sunflower-derived PS, which may differ in efficacy due to different fatty acid profiles. The evidence is strongest for exercise-induced cortisol, with less consistent findings for resting cortisol or chronic psychological stress.
