Resveratrol (3,5,4'-trihydroxy-trans-stilbene) is a naturally occurring polyphenolic stilbene produced by certain plants in response to stress, injury, or fungal infection. It is found in red grape skins, Japanese knotweed, peanuts, blueberries, and red wine. Resveratrol gained significant public attention after studies showed it activated sirtuins (SIRT1) and extended lifespan in yeast, worms, and mice on high-fat diets. However, its extremely poor oral bioavailability in humans (less than 1% reaches systemic circulation unchanged) raises fundamental questions about whether oral supplementation can achieve biologically relevant tissue concentrations.
How It Works
Resveratrol activates SIRT1, a NAD-dependent deacetylase linked to cellular stress resistance and metabolic regulation, mimicking some aspects of caloric restriction. It also activates AMPK signaling, improves mitochondrial function, reduces NF-kB mediated inflammation, and inhibits platelet aggregation. In cell culture and animal models, these mechanisms produce impressive anti-aging, anticancer, and cardioprotective effects. However, rapid hepatic metabolism converts most oral resveratrol to sulfate and glucuronide conjugates within minutes of absorption, making it unclear whether parent resveratrol or its metabolites are responsible for any observed human effects.
