What It Is
Magnesium L-threonate is a patented magnesium salt of L-threonic acid (a metabolite of vitamin C), developed by MIT researchers specifically to increase brain magnesium concentrations. Unlike other magnesium forms (citrate, glycinate, oxide) that primarily raise serum and peripheral tissue magnesium, threonate was shown in animal studies to uniquely elevate cerebrospinal fluid magnesium levels. The compound is marketed under the brand name Magtein. It gained popularity in the nootropic community after the landmark 2010 Neuron paper showed enhanced learning and memory in rodents. However, it provides relatively low elemental magnesium per dose (~8% by weight), making it a poor choice for correcting general magnesium deficiency and positioning it specifically as a brain-targeted supplement.
How It Works
Magnesium L-threonate enhances cognitive function by increasing magnesium concentration in the brain, particularly in the hippocampus and prefrontal cortex. Elevated brain magnesium increases the density and function of NMDA receptors and synaptic connections between neurons (synaptogenesis). It promotes long-term potentiation (LTP) in the hippocampus, the cellular mechanism underlying learning and memory formation. Magnesium also acts as a natural NMDA receptor modulator, blocking the channel at resting membrane potential to prevent excitotoxicity while allowing signal transmission during active learning. The threonate carrier appears to facilitate magnesium transport across the blood-brain barrier more effectively than other magnesium salts, explaining the preferential increase in CNS magnesium levels observed in animal models.
