It serves as a precursor to both androgens and estrogens through peripheral conversion. DHEA levels peak in early adulthood and decline steadily with age, reaching 10-20% of peak values by age 70-80. This age-related decline led to the hypothesis that DHEA supplementation might reverse some aspects of aging, though large clinical trials have largely failed to confirm dramatic anti-aging benefits.
How It Works
DHEA acts primarily as a prohormone, being converted peripherally to more potent androgens (testosterone, DHT) and estrogens (estradiol, estrone) depending on tissue-specific enzyme expression. It also binds to sigma-1 receptors in the brain, potentially affecting mood and neuroprotection. DHEA may directly modulate immune function, enhance insulin sensitivity, and affect vascular endothelial function. In women, supplemental DHEA can meaningfully increase androgen levels (and thus libido) because baseline androgen production is lower. In men, the additional testosterone conversion from DHEA supplementation is typically too small relative to gonadal production to produce significant effects.
